- Synopsis
- Introduces gene technology and demonstrates the laboratory techniques for performing gel electrophoresis and DNA transformation. The video reviews the structure of the DNA molecule and then explains and demonstrates: 1) ‘cutting and pasting’ - how a target sequence of DNA can be cut using restriction enzymes and combined with DNA from another organism using DNA licase to form recombinant DNA; how gene therapy can be used to replace defective genes. 2) Shows in detail the laboratory technique of gel electrophoresis which is used to separate specific segments of DNA and discusses how DNA analysis is used in biological research; 3) why bacteria are used for transferring DNA (smaller genomes, DNA contained in circular plasmids which provide useful vectors for delivering new genes to other cells); the use of marker genes; 4) DNA transformation in the laboratory - demonstrates an experiment to insert recombinant DNA into E.coli bacteria (genes enabling the bacteria to be more antibiotic-resistant and/or glow in the dark); 5) the importance of safety measures and sterilisation techniques when performing biological experiments in the laboratory.
