- Synopsis
- Shows how to construct DNA libraries in vectors, how to subsequently introduce them into bacteria and the screening methods used to isolate the DNA fragments of interest. Explains and compares the construction and use of the two main types of library - genomic and cDNA libraries. The section on genomic libraries covers the use of phage vectors, cosmid vectors, replica platings, radio-labelled probes, long-range mapping and cosmid walking techniques. Poly C tailing, the use of linkers, mRNA probes, chemically synthesised oligonucleotide probes and immunological identification techniques are featured in the section on cDNA libraries.
