Synopsis
Preparation: guinea pig ileum. Describes the isolated organ response recording technique and includes setting up procedures of the mammalian intestine preparation, the principles of isolated organ preservation and the method of recording tension changes. Gives theoretical consideration to agonist receptor interactions together with development of the concept of receptor occupancy and the derivation of the Clark equation. The Biovideograph record includes details of the drug administration cycle and the initial steps in the conditioning of the preparation to ensure response reproducibility and submaximal responsiveness. Shows the increase in response magnitude resulting from administration of a range of agonist doses varying from threshold to maximum concentrations.
