Non-Competitive Antagonism

Synopsis
Preparation: guinea-pig ileum. Non-competitive antagonism can arise from either non-equilibrium receptor blockade or inhibition of an intracellular process distal to the site of agonist receptor action. The Biovideograph section shows the obtaining of log dose response relationships to the agonists histamine and acetylcholine. Non-competitive antagonism is produced as a result of a single incubation of the preparation with phenoxybenzamine. The differential irreversible inactivation of the receptor population is demonstrated by the extent of the reduction in the maximal agonist response magnitude as well as the rightward shift of the log dose response curves to histamine and acetylcholine. The concluding part of the programme contains a commentary on the distinguishing features of competitive and non-competitive antagonism.
