Large-Scale Preparation of Phage Dna
- Synopsis
- Lambda bacteriophage are used to clone large fragments of DNA. Here, phage are isolated by first infecting a liquid suspension of bacteria. During incubation the bacteria are lysed by the phage. The phage are then completely released from the bacteria with chloroform and purified by cesium chloride density centrifugation. Subsequently, the phage are extracted with phenol/chloroform extraction and dialysed with buffer.
